EPINEPHRINE POTENTIATES ANALGESIC AND ANTIDEPRESSIVE ACTION OF AMITRIPTYLINE AS A RESULT OF STIMULATION OF GASTRIC MUCOSA AFFERENTS

Intramuscular administration of amitriptyline in the minimal effective dose causes the maximal analgesic and antidepressive effect, and also considerable sedation in rats. The combined administration of amitriptyline with adrenaline in the threshold, noneffective doses alone, accordingly, 1/10 and 1/30 part from their minimal effective dose, leads to development of the maximal analgesic and antidepressive effect, but does not produce side sedative effect. The basis for mechanism of potentiation of analgesic and antidepressive effects of amitriptyline is the stimulation of gastric vagal afferents by adrenaline.

Hi Sserdyuk.

Thanks for sharing your article with us. Very interesting topic.
My understanding was combined use of Epinephrine with Amitriptyline causes high blood pressure.
Is this approach safe for pt with underlying cardiac condition. Thanks

dear Admin !Thank You for You nice comment . I dont measure arterial pressure in my experiments,but epinephrine and amitriptyline used in very small doses (1\30 -1\10 from effective doses ) else not increase arterial pressure
